TyVAC-Nepal is a participant- and observer-blind, 1:1 individually randomised trial of the typhoid conjugate vaccine (Vi-TCV) that aims to study the impact of Vi-TCC in an endemic setting.

This study is a continuation of the typhoid vaccine study (TyVAC Nepal), which aims to assess the medium-term efficacy of the Typhoid Conjugate Vaccine (TCV) in children by comparing the relative risk of typhoid fever in a TCV cohort initially vaccinated in 2017/2018 with TCV cohorts vaccinated in 2020 and 2021.

The primary objective of the ACT-South Asia was to determine if the combination of azithromycin and cefixime for seven days results in significantly fewer treatment failures when compared with the current standard regimen of azithromycin alone for seven days in the out-patient treatment of children and adults with uncomplicated typhoid fever (infection due to Salmonella Typhi and Salmonella Paratyphi A) in South Asia.

This study will decode the genetics of S. Typhi samples from South Asia obtained from the ACT-South Asia trial to investigate the epidemiological and evolutionary pathways of azithromycin-resistant S. Typhi.
